In 2001, two physicians at a Department of Veterans Affairs medical centre took four magnesium products people were actually buying, gave them to volunteers, and measured how much showed up in their urine — the standard proxy for how much got absorbed.

And it isn't one lonely paper. A decade earlier, researchers at UT Southwestern ran citrate against oxide in volunteers. Over the four hours after the dose, the citrate group's urinary magnesium rose more than thirty times as much.[2] The same team found magnesium oxide “virtually insoluble in water,” and only 43% soluble even in simulated peak stomach acid.[2]

The thing you swallowed barely dissolved. So why is it on every shelf?

The first villain

Magnesium oxide is cheap in bulk and dense — a high percentage of the compound is magnesium by weight. So a manufacturer can put an enormous, impressive-looking number on the front of a bottle for almost nothing.

The big number on the front isn't a promise. It's a prop.

In 1994, a team at the University of Chicago ran a double-blind, randomised crossover trial using isotope-tagged magnesium so they could follow exactly where it went.[3] What they concluded about magnesium diglycinate — magnesium bonded to two glycine amino acids — was that part of it appears to be absorbed intact, probably through a dipeptide transport pathway.

Twelve patients, all of them with intestinal resection. A small study in a specific group, and it tested the ingredient rather than any finished product — so treat it as what it is: the paper that identified the route, not a verdict on any bottle.

The mechanismThe Second Door

Ordinary magnesium salts queue at one entrance: dissolve in stomach acid, then be absorbed as a loose mineral. If the compound doesn't dissolve well — and oxide famously doesn't[2] — the queue barely moves.

A chelate carries its own key. Part of it appears to be taken up as a whole molecule through an amino-acid transport route[3] — a different entrance, one that doesn't depend on the same bottleneck.

Same mineral. Different door.

That's the whole argument for chelated bisglycinate, and it's why the form costs more to make than the one sitting in your drawer.

Word got out that bisglycinate was the good stuff, and “glycinate” started appearing on the front of bottles everywhere. Which brings us to the second villain — the slipperier one, because it's wearing the first one's disguise.
